As a child she displayed a spirited, tomboyish streak; at 13 she was enrolled in Kennedy International Residential School in Vijayawada. Her early interest in performance became clearer during school when she played Julius Caesar in a tenth-standard stage production, a role that helped convince her to pursue acting.

She completed school in Vijayawada and went on to study at Villa Marie Degree College for Women in Hyderabad, earning a BA with a focus on psychology and journalism. Alongside formal education, Punarnavi trained with theatre groups in Hyderabad, including Samahaara and Nishumbita, to develop her acting skills.

Punarnavi made her Telugu film debut in 2013 with Uyyala Jampala, appearing as the character Sunitha. Her mother, a fashion designer, designed her outfit for that first film. Early recognition came quickly: in 2012 she won the Fresh Face of Vijayawada title in a local pageant, and in 2014 she was featured in the magazine You & I.

Following her debut, Punarnavi continued to build a steady filmography in Telugu cinema. Her credits include Malli Malli Idhi Rani Roju, Ee Cinema Superhit Guarantee, Pittagoda, Manasuku Nachindi, Enduko Emo, and Oka Chinna Virama Tharvatha. She has described acting as a path she pursued with confidence, and she attributes parts of her early success to family encouragement and mentors from theatre.

Outside of films, Punarnavi appeared on television reality programming when she took part in Bigg Boss Telugu Season 3 in July 2019. That appearance raised her visibility with a broader Telugu-speaking audience.

In interviews and public comments she has said that if she were not an actress she might have worked as a mental health practitioner, reflecting her academic interest in psychology.
